The situation

What a binary readout leaves unanswered

Vaporised hydrogen peroxide (VPHP) bio-decontamination cycles are conventionally developed and validated using biological indicators (BIs) containing Geobacillus stearothermophilus spores. While BIs remain essential for Performance Qualification and requalification, their binary readout, lot-to-lot variability and the occurrence of rogue or late-positive results provide only limited insight into actual cycle performance.

  • Selecting and ranking true worst-case locations can be challenging in complex isolator systems.
  • BI lot variability and rogue or late-positive results can complicate cycle development, PQ and RQ investigations.
  • Quantitative EI data can improve process understanding, but their role, interpretation and acceptance criteria must be clearly defined before they can be integrated into qualification decisions.

What is an Enzyme Indicator?

An Enzyme Indicator is a rapid, quantitative indicator placed inside the chamber alongside biological indicators during a VPHP bio-decontamination cycle. It gives a quantitative readout of hydrogen peroxide exposure at each position in approximately 60 seconds, with no incubation period.

Do Enzyme Indicators replace biological indicators?

No. Biological indicators remain essential for Performance Qualification and requalification. Enzyme Indicators add rapid, quantitative data during cycle development and investigation. They do not override a positive BI result.

What is a rogue biological indicator?

A rogue or late-positive biological indicator returns a positive result without a corresponding process failure. Rogues are a known phenomenon and, statistically, even a low incidence rate can affect the outcome of a Performance Qualification, particularly where large numbers of indicators are used.

What is VPHP bio-decontamination?

VPHP stands for vaporised hydrogen peroxide. It is the method used to bio-decontaminate isolators, RABS and filling lines in aseptic pharmaceutical manufacturing, and the cycle is validated using biological indicators containing Geobacillus stearothermophilus spores.

Does Annex 1 allow alternative methods to biological indicators?

Annex 1 permits alternative methods where equivalence or superiority is demonstrated, and says rapid and alternative methods and continuous monitoring systems should be considered. Enzyme Indicators complement biological indicators rather than replacing them in qualification.

Is this webinar vendor-specific?
